The reversible binding of half-cystine residues to serum protein, and its bearing on the cystine requirement of cultured mammalian cells.

چکیده
All the serially propagated mammalian cell cultures SO far examined, as well as monkey kidney cells in primary passage, have been found to require a minimum of 13 amino acids (l-4). One of the 5 amino acids regularly required in culture, but not necessary for nitrogen balance in man, was L-cystine. In many of the experiments, however, there was definite if limited initial cellular growth in a cystine-free medium, indicating the presence of a mechanism for its provision. It was subsequently found that a number of inorganic reducing agents containing sulfur (S’, S~0’3, S30;, SO;) would greatly promote the growth of human, monkey, and mouse cells in a presumably cystine-free medium, and that D-cystine, D-homocystine, L-cysteamine or L-cystamine, and thioglycolate were also effective. As will be here shown, these compounds are not used for the biosynthesis of L-cystine, but act by mobilizing half-cystine residues bound through disulfide bonds to the serum protein of the medium.

The Effects of Novel Mutations in A1 Domain of Human Coagulation Factor VIII on Its Secretion Level in Cultured Mammalian Cells
Inefficient secretion of the human coagulation factor (hFVIII) in mammalian expression systems is one ofthe main causes of the hFVIII low expression level, attributed to its interaction with a chaperone known asBiP/GRP78. In order to improve secretion efficiency of the hFVIII, based on the higher secretion level of theporcine FVIII and analysis of the hFVIII A110 region, that ...
متن کاملEvaluation of an Albumin-Binding Domain Protein Fused to Recombinant Human IL-2 and Its Effects on the Bioactivity and Serum Half-Life of the Cytokine
Background: Cancer immunotherapy is a promising strategy for cancer treatment. In this strategy, the immune system is triggered to destroy cancer cells. IL-2 is an important factor in passive cancer immunotherapy that helps modulating some important immune functions. One of the IL-2 limitations is low serum half-life; therefore, repetitive high doses of the injections are required t...
متن کاملStructure of an antifreeze polypeptide from the sea raven. Disulfide bonds and similarity to lectin-binding proteins.
The antifreeze polypeptide (AFP) from the sea raven, Hemitripterus americanus, is a member of the cystine-rich class of blood antifreeze proteins which enable survival of certain fishes at sub-zero temperatures. Sea raven AFP contains 129 residues with 10 half-cystine residues. We have analyzed these half-cystine residues and established that all 10 of the half-cystine residues appeared to be i...
متن کاملEvolutionary Analysis of Mammalian ACE2 and the Key Residues Involved in Binding to the Spike Protein Revealed Potential SARS-CoV-2 Hosts
Introduction: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spilled over to humans via wild mammals, entering the host cell using angiotensin-converting enzyme 2 (ACE2) as receptor through Spike (S) protein binding. While SARS-CoV-2 became fully adapted to humans and globally spread, some mammal species were infected back. The present study evaluated the potential risk of mammals...
